Naphthalimide derivatives have multiple biological activities, including antitumour and anti-inflammatory activities. We previously synthesized several naphthalimide derivatives; of them, compound 5 was found to exert the strongest inhibitory effect on human DNA topoisomerase II activity. However, the effects of naphthalimide derivatives on platelet activation have not yet been investigated. Therefore, the mechanism underlying the antiplatelet activity of compound 5 was determined in this study. The data revealed that compound 5 (5–10 μM) inhibited collagen- and convulxin- but not thrombin- or U46619-mediated platelet aggregation, suggesting that compound 5 is more sensitive to the inhibition of glycoprotein VI (GPVI) signalling. Indeed, compound 5 could inhibit the phosphorylation of signalling molecules downstream of GPVI, followed by the inhibition of calcium mobilization, granule release and GPIIb/IIIa activation. Moreover, compound 5 prevented pulmonary embolism and prolonged the occlusion time, but tended to prolong the bleeding time, indicating that it can prevent thrombus formation but may increase bleeding risk. This study is the first to demonstrate that the naphthalimide derivative compound 5 exerts antiplatelet and antithrombotic effects. Future studies should modify compound 5 to synthesize more potent and efficient antiplatelet agents while minimizing bleeding risk, which may offer a therapeutic potential for cardiovascular diseases. 相似文献

Appressorium formation in germinating conidia of Magnaporthe grisea was inhibited on inductive and on noninductive surfaces by monounsaturated fatty acids with chain lengths of 16, 18, or 20 carbon atoms. On a noninductive surface, the inhibition was only observed upon stimulation with 1,16-hexadecanediol or oleyl alcohol, but not upon stimulation with 8-(4-chlorophenylthio)-adenosine-3′,5′-monophosphate. In the C18-series, fatty acids with a double bond in position 9 were the most active ones. At 1 μg/ml of oleic or elaidic acid, less than 30% of the germinated conidia formed appressoria. The mode of inhibition was competitive to the inducing agent. On an inductive surface, compared to a noninductive surface the concentrations of oleic and elaidic acid needed for inhibition of appressorium formation were one order of magnitude higher. Methyl esters of inhibitory fatty acids and acids with two double bonds were not active. Like oleyl alcohol, elaidyl alcohol and petroselinyl alcohol stimulated infection structure formation on the noninductive surface. 相似文献

Abstract In order to test the hypothesis that arthropod-induced neoplastic formations on trees affect biochemical characteristics of both the newly formed galls and host plant tissues, biochemical characteristics with a possible adaptive role were determined in nine gall-former–host tree combinations. Photosynthetic pigments, extractable protein content, and oxidative enzyme activities were determined in gall tissues, leaf tissues of galled leaves, and leaves on ungalled tree branches. Neoplastic tissues were characterized by a low content of photosynthetic pigments, decreased chlorophyll a/b ratio, lower extractable protein content, and decreased activities of peroxidase and polyphenol oxidase as compared with ungalled host leaf tissues. In galled leaves or in leaves adjacent to galls, increased level of peroxidase activity was found. In several gall-inducer–host plant combinations, galled host plant tissues contained increased activity of polyphenol oxidase as well. The presented data reflect long-term systemic effects of neoplastic formation on host tree physiology suggesting that gall inducers affect potential adaptive responses of host plants. 相似文献

This article analyses the Urban Poverty and Family Life Survey of Chicago and an urban subsample of the National Survey of Families and Households to examine race and ethnic variation in the occurrence and consequences of birth versus marriage pathways to family formation. Results based on multiple‐decrement life table analysis and multi‐variate life table regression analysis reveal strong race and poverty effects on pathways to family formation, but Hispanic origin does not systematically influence the odds that women will enter family life via marriage versus birth. We show stronger race effects on pathways to family formation for the Chicago sample owing to the much higher incidence of teen parenting among black inner‐city residents. Results also suggest lasting economic consequences of non‐marital fertility, irrespective of whether women eventually marry or divorce subsequent to a premarital birth. 相似文献

Adventitious shoots were induced from excised embryos of Pinus resinosa Ait, on half-strength Le-Poivre (LP) medium containing 1–70 μ M N6 -benzyladenine (BA). At lower concentrations of BA, only 2–3 shoot primordia (from as many as 22 formed per embryo) developed into shoots when subcultured onto medium containing 0.5% activated charcoal. Concentrations of 10 to 70 μ M of BA produced significantly higher numbers of shoot primordia and most of them developed into shoots. Ten to 17 day culture on medium containing 10–25 μ M BA proved optimal for maximum adventitious shoot production. Less than three days of incubation on the cytokinin medium did not stimulate the formation of adventitious shoots. Twenty-four day culture on the same medium produced several shoots, but most of them failed to develop normally and formed callus. Coconut milk (0.1–5% v/v) inhibited adventitious shoot formation. Using optimal conditions, seeds from 11 open-pollinated selected trees were compared to test for genetic differences in the potential to produce adventitious shoots from embryos. No significant differences were observed with regard to the shoots produced per embryo among the different seed collections. More than 200 plants produced through this technique were tested for variation in several isozymes by electrophoresis. No variations were observed. 相似文献

The deme-formation hypothesis states that selection can produce adaptive genetic variation within and among phytophagous insect populations. We conducted three field experiments and tested this prediction by transferring eggs and measuring performance of a mobile leafmining insect, Stilbosis quadricustatella. In Experiment 1, we compared the rate of mine initiation of leafminers transferred to natal and novel sites. In Experiment 2, we compared mine-initiation rate of leafminers transferred to natal and novel host-plant species. In Experiment 3, we compared the mine-initiation rate, mine-completion rate, and sources of mortality of miners transferred to neighboring natal and novel Quercus geminata trees. In the first, second, and third experiments, leafminer larvae initiated significantly more mines at the natal site, on the natal plant species, and on the natal Q. geminata tree, evidence for adaptive differentiation. Furthermore, plant-mediated mortality was significantly lower among miners transferred to natal Q. geminata trees. This result supports a key assumption of the deme-formation hypothesis: insects adapt to the defensive phenotypes of individual trees. However, natural-enemy mortality was significantly higher among miners transferred to natal trees, essentially reversing the plant effect. Therefore, rates of successful mine completion were similar on natal (19%) and novel (17%) trees. This experiment suggests that host plants and natural enemies may represent opposing forces of selection. Leafminers adapted to individual trees may realize a selective advantage only when natural-enemy densities are low. 相似文献

Dispersed from a dry film into buffer (5 mM phosphate, 0.15 M NaCl, pH 7.4), the liponucleotide 1-β-d-arabinofuranosylcytosine 5′-diphosphate l-1,2-diacylglycerol (ara-CDPdiacylglycerol) spontaneously forms vesicles which are several microns in diameter and probably unilamellar. Their average size immediately begins to decrease, and after 2 h none can be seen in the light microscope. During 1–2 days in unstirred solutions at 25°C, the vesicles are transformed to spherical or nearly spherical micelles having an apparent partial specific volume of 0.835 ml·g?1, a maximum possible aggregation number of about 150, and an anhydrous radius of about 37 Å. The critical micelle concentration (CMC) is about 10 μM in buffer and 20 μM in distilled water, but micelle-monomer equilibration requires at least 1 week at a total concentration of 66 μM. This exceedingly slow equilibration is unique among reported detergents. The standard enthalpy and entropy of micellization are ?13 kJ·mol?1 and 87 J·mol?1·K?1, respectively. These values are within the range reported for other detergents. Sonication accelerates the vesicle-micelle transformation to 30 min. 相似文献
